One of the up strokes will be compression, the other will be the exhaust stroke and will have the exhaust valve open, (much lower feel of compression on your thumb)

Another easy way to verify you are TDC on compression stroke #1 cylinder is to see what tower on the distributor goes to #1 spark plug. Take the distributor cap off and make sure the rotor is pointing to that plug wire tower.
