Timing a 318 v/8 in a 1965 w-300

I just bought a w-300 1965 with the 318 v/8
and I need to know which cylinder is number one for timing purposes. Also am I better off leaving the 2 barrel carb on it or better off with a 4 barrel and manifold? Should I set timming at 12.5 BTDC ? Thanks for any help. I bought the truck on E-Bay from a fire dept in Maryland.
