Considering the four positions forward of neutral, count from the left, 1, 2, 3.... to the third position. That will be 5th if it is an overdrive five speed. Put the transmission in that position, then rotate the input shaft one turn, counting the revolutions of the outpput shaft. If it is an overdrive, the output shaft will turn more than one revolution.

If it is a fifth direct transmission, the output shaft will turn slower than the input shaft.

The right-hand position on the rear part of the pattern will always be direct, and that is in both cases, meaning it will be 5th gear in a fifth direct transmission and it will be 4th in an overdrive. So, if you are used to a fifth direct NP 54X trans, don't just put it into that position, determine that to be direct, and reject it because you believe it to be a fifth direct transmission.
