Easiest and arguably best way to get what you want is to either put the bus body on a modern chassis, or swap the drivetrain under the bus frame.

That way you are starting with an entire drivetrain that was engineered and designed for the types of speeds you want to do. You can adapt everything to the bus frame but that's a ton of work and you will be responsible for trying to keep all the parts in the right position and alignment.

You will loose some of the look, and there is still a lot of work in adapting the body to the frame and controls to the body.

It's not a very Dodge answer but if you are really stuck on Diesel with a vehicle that heavy a Ford or Chevy V8 might be a better fit under the short hood while still making the kind of power you are probably looking for. A Duramax with the 6-speed Alison would make a durable drivetrain and be pretty easily supported by the aftermarket.
