It shouldn't be a big deal to check valve timing and spark timing and make sure it's right. You need to start at the beginning and make sure the basics are correct before proceeding.

If it's right and you have compression, fuel (which should be adequately distributed to all cylinders using a carb or TBI) and spark then the next logical conclusion would be inadequate fuel to that one cylinder either by blockage in the runner or by a vaccuum leak.

You can check for a vacuum leak by spraying carburetor cleaner (yes, yes, yes!, I know it can be dangerous, but, so can waking up each morning)around the vaccuum fittings on the carb and on the intake manifold runner to the cylinder in question. If the engine responds, then you have found at least one problem.
