Yes it is a check valve. Typically there should only be vacuum flow when the booster/ brakes are in use unless the leak is upstream of the valve. You might be hearing the charcoal canister purge valve in operation. It hangs on a stand alone bracket near the booster with a vacuum line running to the intake and another that disappears down to the canister. If the booster or the diaphragm truly did have a leak you can usually hear it for several seconds after the engine is shut off.
