This sure sounds like a timing problem with the backfire, although the starting fluid can certainly do that as well. I agree with checking the distributer as it does have teeth and being off by a few teeth can make it so you cannot advance/retard your distributer far enough for it to start. If you find that the distributor rotor is actually truly at cylinder ones cap post, you may want to try to rotate it 180 degrees. If you really want to do it correctly, you will need to pull the head if it is an L-6 and get a look at the valves to make sure that when the engine is at TDC the intake valve opened then closed and you are now at TDC. This should be spark time at cylinder one. If this looks ok, I would cycle the engine several times and then look at it again to make sure you are not missing one tooth, causing the timing to "walk" one tooth each rev. If all this fails, you will have to pull off the timing cover and just start from scratch. Before all of this, I would put a spark plug in each wire and make sure all cylinders are getting spark and not just one or two.
