The Hole should indeed be plugged. It is the oil level plug, when oil runs out the winch is "full". The manual just calls it a "plug", I assume it is pipe threads but don't know for sure. If that has been missing or broken for awhile it could have allowed moisture into the housing. The grease fitting you ask about is the yoke fitting where the shear pin resides. It shouldn't take much grease, only enough to keep the joint from freezing up and allowing easier disassembly is the only purpose that I see. Again, maybe somebody else can add more info...
