Tim,

Running them in Parallel:
The Radiator Cooler will help warm the Tranny Fuel up in the winter so that the Tranny will start working better quicker, but also only cool the Tranny fluid down to the coolest temperature of the Radiator.
The Tranny fluid will always take the path of least resistance, so if the additional cooler has a higher resistance to fluid flow than the one in the Radiator, the more of the fluid will pass through the Radiator than the extra cooler, reducing its cooling effect.

Running them in Series:
As long as you pass the fluid through the Radiator first it can pre-cool the fluid reducing the work on the additional cooler potentially cooling the tranny fluid even lower than either cooler buy them selves.
This does add both coolers fluid resistance to the flow of the tranny fluid meaning less fluid cooled.
